Investigators found that 90% of the reports of self-injurious behavior or depression were associated with the use of varenicline. Bupropion and nicotine replacement products were responsible for the remaining 7% and 3%, respectively.
The FDA concludes: "Moore et al's conclusion that 'varenicline shows a substantial, statistically significant increased risk of reported depression and suicidal/self-injurious behavior' is misleading, as [proportional reporting ratios] are measures of association and not measures of risk,".
